该剧以19世纪80年代到1927年密西西比河上一条演艺船为故事背景，讲述了长达半个世纪之久人世的悲欢离合，具有史诗一样的气魄。全剧分为两幕，第一慕开始于“棉花盛开”（Cotton Blossom）号演艺船来到密西西比河畔那彻斯（Natchez）码头，船长安迪的女儿，单纯的麦戈诺丽娅（Magnolia）与相貌英俊的流浪赌徒拉维纳尔（Ravenal）一见钟情（唱《请相信》（Make Blieve）），黑人船工乔（Joe）注意到他们之间的感情，又想到自己的身世，唱起《老人河》。此间，剧团的工程师皮特因为向演艺船上的明星茱莉（Julie）小姐献殷情，被茱莉的丈夫斯蒂夫打伤，皮特为了报复，揭发出茱莉有黑人血统，这在当时触犯了禁止不同种族通婚的法律。市长赶来逮捕斯蒂夫和茱莉，斯蒂夫划破茱莉的手，喝了茱莉的血。他问市长是否白人身上有黑人的血，他就是个黑人，市长回答，谁只要一滴黑人的血，他就是黑人。斯蒂夫说他现在身上有不止一滴黑人的血，所以也是黑人，并没有违反不同种族禁婚的法律。市长无言以对，于是下令禁止 “棉花盛开”号晚上的演出。斯蒂夫和茱莉为避免拖累剧团，离开了演艺船。麦戈诺丽娅和拉维纳尔代替茱莉和斯蒂夫登台表演。当晚他们表白了爱慕之情（唱《你就是爱》（You Are Love））。第一慕结束于麦戈诺丽娅和拉维纳尔的婚礼。
第二幕开始于麦戈诺丽娅与拉维纳尔的女儿吉姆降生。拉维纳尔一家迁到了芝加哥，靠拉维纳尔的赌博过活，不久破产，全家被赶出了旅店。拉维纳尔抛弃了妻子和女儿吉姆，离家出走。麦戈诺丽娅在一家夜总会找工作时遇到了已与斯蒂夫分手的茱莉，茱莉把自己的工作让给了她。第二天晚上，也就是1899年的圣诞夜，麦戈诺丽娅在演出时与父母相见，她的演出获得了成功。二十多年之后，麦戈诺丽娅成为音乐喜剧明星。第一次世界大战之后，茱莉沦为乞丐。1927年，安迪写信邀请拉维纳尔作客演艺船，同时招来了麦戈诺丽娅和她的女儿吉姆，一家人终得团圆，已经成了百老汇的明星的吉姆表演了节目。全剧终于乔再次唱起《老人河》，这首歌是交织情节的关键，也是点睛之笔：
