故事发生在陕西，农民刘高兴（郭涛 饰）是个喜欢设计飞行器的有心人。迫于生计，他跟好兄弟五富（冯砾 饰）一起来到西安打工，在老乡韩大宝（林雪 饰）的一间破旧仓库落脚，邻居包括东北大汉黄八（海一天 饰）和一对夫妇种猪（巴多 饰）和杏胡（苗圃 饰）。高兴的工作是收废品，虽然工作不那么光鲜，但是高兴却乐在其中，这种积极的心态感染了五富和邻居夫妇。另外，碰到小流氓石热闹（黄渤 饰）寻衅滋事，也是高兴挺身而出惩恶扬善。

An intense drama on the portrayal of brotherhood, BROTHERS depicts the mistrust and feud between two brothers through the eyes of an investigating police officer. Despite their close relationship, Yiu and Shun were separated at a young age due to a decision made by their father. Years later they meet again when the family runs into crisis where Yiu, inspired by his father's last words, manipulates a series of setups which endangers Shun's life, so as to achieve his goal. The crisis brings the two brothers back together again. However, a series of setups arranged by Yiu causes Shun to flee to Thailand, accompanied by confidantes Ching (Crystal Huang) and Ghostie (Wong Yat-wah). As Shun is caught between life and death, the trust between the brothers is put to a serious test. When Shun finally realizes the true intention of his brother, things are already beyond repair.

ICAC principal investigator William Luk takes his job seriously, and he needs every last scintilla of support from his team to take on the powerful and cunning Supt Wong, who is corrupt to the core. Bumbling private detective Chan Tam is enlisted by police pal Fung Chak to help in the investigation of a serial murder case. The victims a middle-aged man killed at home, a dead woman found in the trash dump, and a teenage girl slain in the park were all killed in grisly manners, but they didn't seem related to one another. With no clue to follow up on, Tam is decidedly at his wits' end.

冰（杨千嬅 饰）是广告公司的客户经理，公司她是女强人，下班后她跟很多女人一样，苦苦寻觅自己的爱情。一天晚上她在酒吧遇到了心理医生V仔（任贤齐 饰），两个人缠绵的时候V仔意外发现了冰的胸部有肿块。经医生诊断证实冰真的患上了乳癌，冰大受打击，惧怕自己因为失去了一边乳房而没有好归宿。
